Effect of Gabapentin on Intraocular Oxidative Stress After Spinal Cord Ischemia-Reperfusion Injury

Materials and Methods: Twenty-four adult, male, New Zealand rabbits were divided into 4 groups; control (n=6), ischemia (n=6), low dose gabapentin (30 mg/kg), (n=6) and high dose gabapentin (200 mg/kg), (n=6) treatment groups. In ischemia and treatment groups, the abdominal aorta was clamped for 30 minutes by an aneurysm clip, approximately 1 cm below the renal artery and 1 cm above the iliac bifurcation. In the control group, only laparotomy was performed in the control group. The animals were killed 48 h later. Vitreous samples were collected for analysis. Levels of tumor necrosis factor-α, malondialdehyde, and total antioxidant status were analyzed as markers of oxidative stress.
Results: Oxidative stress is observed to increase in vitreous following spinal cord ischemia-reperfusion injury. In ischemia group the vitreous malondialdehyde and tumor necrosis factor-α levels were significantly higher (p<0.05) and total antioxidant status level was significantly lower compared to the conrol group (p<0.05). The vitreous malondialdehyde and tumor necrosis factor-α levels were significantly lower (p<0.05) and total antioxidant status significantly higher in low and high dose gabapentin groups compared to the ischemia group (p<0.05). When low and high dose gabapentin groups compared with control group, no significant difference were found between vitreous malondialdehyde, tumor necrosis factor-α, and total antioxidant status levels (p>0.05).
Conclusions: The experiments conducted in the present study show that gabapentin has a significant effects on lowering the oxidative stress in the eyes following a spinal cord ischemia-reperfusion injury. Keywords : Gabapentin, oksidatif stress, spinal cord injury, tumor necrosis factor-α, vitreous
